A city on the resource will allow you access to the resource. You don't have to demolish the city to get it. You simply lose out on the extra productivity the resource + improvement adds to the tile.
For example, if you build a city on an iron resource, you will have access to swordsman and other units you can build from iron. However, it's not a very efficient thing to do, because a mine on an iron tile creates a HUGE boots to hammers the tile provides, while a city on the tile will receive at maximum a +1 boost.
Though you should keep in mind that often later resources are revealed before you can actually make use of them. In the case of oil, for example, it is revealed by the Scientific Method technology, but you cannot actually build a well to gain access to the resource until you have the Combustion technology (or Plastics for water-based oil). Ergo, if you build a city on an oil resource, you won't be able to use the oil until Combustion, even if you reveal it when you discover Scientific Method.
