Repeaters are easy to build, they just require bars of the appropriate material. Alternately, you could buy a shotgun from the arms dealer or farm the Wall of Flesh for a clockwork assault rifle, as crystal bullets should be fairly effective against the Destroyer. I still think repeaters are better because of archery potions, and unholy arrows, holy arrows, jester's arrows, and hellfire arrows are all pretty effective against the Destroyer. The caveats for each of the arrows are as follows:
Unholy Arrow: The easiest to obtain, but also probably slightly weaker than the other options. You need good aim to use them to their greatest effectiveness, firing so it pierces multiple sections. Since they can pierce up to 5 times they are generally superior to meteor shot against the Destroyer.
Jester's Arrow: Impossible to craft, you must find them. Thus you probably have a limited supply, far fewer than you'd need for the Destroyer and its 80,000 HP. Otherwise their ability to pierce infinitely with good damage makes them an excellent (but impractical) choice.
Holy Arrows: Better damage than the unholy arrow (lower base but the stars pierce infinitely and have good damage) and much easier to aim, but an absolute pain to obtain, as you'll need around 1500 to be safe and it takes 1 unicorn horn per 35. Unicorns are fairly uncommon spawns.
Hellfire Arrows: About as easy to obtain as unholy arrow and about as effective in "piercing," but easier to aim and with better damage. The splash damage will damage multiple sections if aimed properly. The main drawback is their splash damage will also damage you if you're too close to the impact point.
Meteor Shot: Fairly effective but since they can only pierce one target I do not recommend them.
Crystal Shot: Has a splash damage effect similar to the Hellfire Arrow, but does not damage you for being too close. A perfectly serviceable alternative with the shotgun or clockwork assault rifle, as they are easier to aim than unholy arrows and will use far less ammunition, yet are still pretty easy to obtain (crystals are everywhere in the Underground Hallow and you don't need very many, a stack of crystals will last you a very long time). The main drawback is that they will do a bit less damage than a repeater due to the lack of a potion that boosts the damage of guns.
I actually do not recommend using a megashark for the destroyer even if you have one (for subsequent kills), as the rapid fire isn't as valuable here as high burst damage. I think you'll prefer the ammo-conserving qualities of the shotgun or clockwork rifle. With that said, the megashark should be your goal to obtain from the Destroyer because you'll need it to fight Skeletron Prime and the Twins.
